Protocol: Prepare the infiltration buffer and adjust pH. Add the required volume of Agrobacterium culture to the buffer. Incubate for 1-2 hours in the dark. Immediately before infiltration, combine the buffer/buffer's based on number of constructs. Finally, add the pre-infiltration additives, mix well and perform vacuum/syringe infiltration.
